Similarity : the DNA is supercoiled in both eukaryotic and prokaryotic gene organisation. 

Difference: 

1) Nucleosome(146bp + histone octamer) is PRESENT in eukaryotes.

Nucleosome is ABSENT in prokaryotes. 

2) Chromatid structures are present in eukaryotes. 

Chromatid structures are absent in prokaryotes. 

3) DNA is present in defined nucleus in eukaryotes. 

DNA is not well defined,  the place where it is found is called nuclein.
